Time-dependent Expression of Cadherin-11 and Col-II in the Temporomandibular Joints of Guinea Pigs With Spontaneous Osteoarthritis
Objectives: Cadherin-11 plays a critical role in evoking synovial fibroblasts (SFs) inflammatory factors that may contribute to synovial inflammation and cartilage degradation. The objective of present study was toinvestigatethe spatial and temporal changes of cadherine-11(CAD-11) and collagen-II(Col-II) in the temporomandibular joint (TMJ) of guinea pigs with spontaneous osteoarthritis (OA). Methods: Dunkin-Hartley (DH) strain guinea pigs and Bristol Strain 2 (BS2) guinea pigsin the age of 1-, 3-, 6-, 9-, and 12 months were analyzed. The condyle of temporomandibular joints of both DH and BS2 guinea pigs were harvested and fixed. Histological micrographs of the condyle cartilage were obtained. The distribution and expression of CAD-11 and Col-II were evaluated through immunohistochemical staining. Results: Cartilage degeneration in TMJ was not found in BS2 guinea pigs, but it can be found in DH guinea pigs after 6 months. Compared with BS2 guinea pigs, increased thickness of the mineralized layer and obvious cartilage destruction can be observed in the TMJ of DH guineapigs from 6 months. Expression of cadherin-11 located in proliferative and transitive layer in 3 months, and then spread to superficial layer in 9 and 12 months, suggesting activation of synovial inflammation is related to the development of OA. Moreover, Col-II–positive signal can be found in the proliferative, hypertrophic,and mineralized layers, which were elevated over time. Conclusions: Our preliminary results suggested that enhanced expression of CAD-11 might contribute to pathogenic process of OA in TMJ at the early stage.
