Identification of Intrinsic Cisplatin Resistance Gene

Objectives: Our aim is to identify anti-cancer drug genes for predicting the chemosensitivity before treatment. Anti-cancer drug resistance genes are divided into intrinsic resistance genes that highly express in resistant cell de novo, and acquired resistance genes that are induced by exposure of anti-cancer drug. Intrinsic resistance genes are thought to be more valuable to predict the sensitivity of anti-cancer drug at the primary clinical onset, although both intrinsic and acquired resistant genes are involved in drug-sensitivity. Now we established new approach to identify not acquired but intrinsic resistance genes.

Methods: We established several cell lines by using single cloning method. The chemosensitivity and gene expression profiles of all cell lines were investigated by using cell survival assay and microarray respectively. We estimated and identified intrinsic genes which expression level correlated with the degree of resistance.

Results: Several genes including Osteopontin and PTP4A1 were identified as intrinsic resistance genes to cisplatin. Osteopontin and PTP4A1 were shown to be related with response of cisplatin by using overexpression technique in vitro due to increasing AKT activation.

Conclusion: These results indicated that this method is suitable to identify the intrinsic resistance genes and these genes may be useful for predicting the response against anti-cancer drug.
