Comparative Cytotoxicity of S. persica , Essential Oil and Chlorhexidine  Mouthwashes

Chewing sticks of Salvadora persica (miswak) have been used as an oral hygiene aid. Mouthwashes are used as antiseptics for plaque control, preventing wound infection, and accelerating healing process. Currently limited information is available on the cytotoxic effects of miswak extract, Essential oil (EO) and Chlorhexidine (CHX) on fibroblast, osteoblasts and macrophages.

Objective:  To compare the cytotoxicity of miswak extract, EO containing mouthwash and CHX mouthwash  on cultured mouse fibroblasts, osteoblast and macrophages. 

Method:

The miswak extract was prepared by soaking 10g of small pieces of miswak into 100 ml of dH2O for 72 hrs. The extract was centrifuged at 5000 rpm for 10 mins, filtered, and prepared in 50% (v/v) concentrations. The miswak extract, EO and CHX were tested in original strength, 1/2, 1/4, 1/8, and 1/16 dilutions.  L929 , RAW 264.7 and MC3T3 cells were seeded into 96-well plates at 3x104 cells/well and incubated with 100 µL cell culture medium for 24 hours to allow cell adhesion. Then 10 µL of tested solution was added to the culture wells.  In the control group, 10 µL water was added.  After 24 hours’ incubation, cell cytotoxicity was evaluated by MTT assay.  Cell viability was calculated as the percentage of the control group. The results were analyzed with one-way ANOVA. 

Result:  

In cultures of fibroblasts, osteoblasts and macrophages, miswak extract had significantly high cell viability than EO and CHX in the original solution (p < 0.05).  Miswak and EO had significant high cell viability than CHX in the ½ and ¼ dilutions (p < 0.05)  and in macrophages, Miswak and EO had significant high cell viability than CHX in the 1/8 dilution (p < 0.05).

Conclusion:

Miswak extract has less cytotoxicity than EO and CHX in original solution. CHX is more cytotoxic than miswak and EO to fibroblasts, osteoblasts and macrophages.
