Fennel essential oil inhibits virulence of Streptococcus mutans

Objective: Dental caries is the most common chronic disease in the dental fields. S. mutans is the most important bacteria in the formation of dental plaque and dental caries. In the present study, we investigated the inhibitory effect of Fennel (Foeniculum vulgare) essential oil on S. mutans biofilm formation and virulence factor expression.

Method: The inhibitory effects of Fennel essential oil on the growth, acid production, biofilm formation and adhesion were investigated in S. mutans. The biofilm formation of S. mutans was determined by scanning electron microscopy (SEM) and safranin staining. Real-time PCR analysis was performed to evaluate the influence of the Fennel essential oil on the gene expressions of virulence factors in S. mutans.

Result: The Fennel essential oil showed concentration dependent inhibitory activity on the growth of S. mutans and significant inhibition of acid production at the concentrations of 0.025, 0.05, 0.1 and 0.2 mg/ml compared to the control group. Furthermore, the biofilm formation was significantly inhibited when the bacteria were treated with Fennel essential oil. Real-time PCR analysis showed that the expression of gtfB, gtfC, gtfD, spaP, gbpB, relA ,brpA, virR  was decreased in S. mutans when it was treated with Fennel essential oil.

Conclusion: These results suggest that Fennel essential oil may inhibit S. mutans biofilm formation and virulence factor expressions.
