MUC1/SEC and Xerostomia in Sjögren's Syndrome Patients

Introduction. Sjögren’s syndrome (SS) patients usually manifest xerostomia (mouth dryness). Mucin type 1 (MUC1) is a large heavily O-glycosylated protein usually localized at apical surface of epithelial cells and participates in hydration of mucosa cell surfaces. MUC1/SEC, a secreted form of MUC1, contains an 11 amino acid peptide named immune enhancing peptide (IEP) at its COOH terminus. This unique peptide in the secreted MUC1 is able to enhance immune system response by up-regulating monocyte chemotactic protein 1 (MCP-1), through pro-inflammatory cytokines  gamma interferon (IFN-g), which play a critical role during the immune phase of Sjögren Syndrome, inducing perpetuation of inflammation in salivary glands (SG).

Objective. Our study determined the MUC1/SEC mRNA and protein levels and its subcellular localization in SG.

Methods. SG biopsies of 16 SS-patients and 16 controls were obtained. Relative MUC1/Sec mRNA levels were determined by qPCR, protein levels by western-blot and immunochemistry for its localization.

 

Results. Relative MUC1/Sec mRNA levels were significantly increased and cytoplasmic protein accumulation was observed in SS-patients.

Conclusion. MUC1/SEC overexpression could explain mouth dryness in SS-patients.  Fondecyt 1080006 (MJG, SA y CM).
