ORAOV1 inhibits tumor metastasis and progression in OSCC

Objective: This study aimed to investigate the biological function and therapeutic effect of ORAOV1 in oral squamous cell carcinomas (OSCC) via both in vitro and in vivo approaches. Method: Short hairpin RNA (shRNA) targeting ORAOV1 was cloned into pGenesil-2 plasmid vector and then transfected into OSCC cells to inhibit the expression of ORAOV1. In vitro, cell growth assay, cell adhesion assay, cell migration assay and cell invasion assay were performed. In vivo, nude mice bearing HSC3 cells were treated twice weekly with intratumoral administration of 50 ug PGEN-shORAOV1 /250 ug liposome complexes (100 ul) for 3 weeks. Tumor volume, toxic reaction and metastasis were observed through the experiment. TUNEL assay, CD34 vessel staining and immunohistochemistry assay of metastasis correlated protein were further conducted on tumor tissue.Result: Our data demonstrated that the shRNA targeting ORAOV1 markedly suppressed the cell growth, cell adhesion, cell migration and cell invasion in vitro, moreover, inhibited the tumor growth and tumor metastasis in vivo, without obvious adverse effect. Furthermore, we found that the down-regulation of ORAOV1 in vivo could increase the tumor angiogenesis and apoptosis, and inhibit the expression of VimentinAMMP2ARhoC in OSCC cells. Conclusion: Taken together, our findings strongly suggest that down-regulation of ORAOV1 inhibit the metastasis and progression of OSCC. ORAOV1 may be a novel target that could be explored to develop therapeutic strategy in OSCC management, especially in inhibiting and preventing tumor metastasis.
