Titanium surface nanocoatings with plant-derived molecules: pectins

Objective: A new strategy to enhance bone healing on implant sites is to develop a surface which will increase the attachment of osteoblasts. Nanocoatings with organic molecules as proteins and polysaccharides change the chemical and physical properties of the surface and there, by influence, cell adhesion, proliferation and differentiation. In the effort to find new coating materials for medical devices, pectin and especially rhamnogalacturonan I (RG-I) have been suggested. The aim of this project was to characterize chemical and physical properties of titanium surfaces nanocoated with different structures of RG-Is.

Method: RG-I was extracted from the destarched potato pulp and the RG-Is structure was enzymatically modified. The RG-Is were characterized with chromatographic techniques. Native and modified RG-Is were covalently coated on titanium surfaces and the nanocoatings were analyzed with immunofluorescence labeling and atomic force microscopy. The titanium surface properties were evaluated by contact angle measurements, atomic force microscope (AFM), and X-ray photoelectron spectroscopy (XPS).

Result: The enzymatic treatment resulted in a reduction of the RG-Is side chains. XPS analysis proved the presence of RG-Is on the coated surface. The titanium surfaces coated with RG-Is became more hydrophilic. The RG-Is nanocoating did not significantly affect the surface roughness, as the AFM studies showed that the RG-Is molecules had an average length of 100 nm and height of 6 nm.

Conclusion: It can be concluded that pectin nanocoatings have an influence on the chemical and physical properties of titanium surface making it more hydrophilic without changing surface roughness.
