Comparison of Fluoride Regimens on Fluorine Uptake in Carious Enamel

Objectives: The purpose of this study was to compare using the fluoride-containing dentifrice (FD) with and without fluoride mouth rinse (FM) on fluorine (F) uptake in carious enamel during pH-cycling by an in-air micro-PIGE/PIXE system.

Methods: Six extracted human teeth were used. Three sections of 170µm thickness were obtained from each tooth. These teeth were divided into three groups; Group A simulated to FD (apply to 1000ppmF twice per day) without FM. Group B simulated to FD with FM (apply to 225ppmF once per day; 5days per week). Group C simulated to no fluoride regimen as a control. The pH-cycling (pH6.8-4.5) was carried out for 5 weeks for preparing artificial carious enamel using an automatic pH cycling system. After pH-cycling, F and calcium distributions of the outer enamel in each specimen were evaluated using micro-PIGE/PIXE system at TIARA (Takasaki Ion Accelerators for Advanced Radiation Application, Takasaki JAPAN). For the comparison of F uptake, the average F concentration in each specimen was calculated at area of 150 µm from the surface. All specimens were taken transverse microradiograph images (TMR) and integrated mineral loss (IML) was calculated from TMR. F uptake and IMLwere analyzed by Games-Howell test, and one-way ANOVA and Scheffe's test, respectively (p<0.05).

For F uptake and IML, there was no difference between group A and B. Group C showed significantly low F uptake or high IML value comparing to that for group A and B.

Conclusion: There was no difference in fluorine uptake between with and without fluoride mouth rinse. Therefore the toothbrush with fluoride-containing dentifrice routine using would be recommended for caries prevention primarily.
