The expression and function of hnRNP A1 in OSCC

Objective: Alternative splicing of pre-mRNA plays important roles in the initiation and progress of oral cancer. The purpose of this study is to understand the expression and function of splicing factor hnRNP A1 in oral squamous cell carcinoma (OSCC).

Method: The expression of hnRNP A1 is analyzed in a tissue array, including two groups: 49 OSCC cases and 10 normal oral mucosal tissues, by immunohistochemistry. The score of each case was calculated by multiplying the percent of cells staining by the intensity score in tumor or nomal tissues. Kruskal-Wallis test was used to analyze the difference of scores between tumor and normal tissues.  OSCC cell line Cal27 cells were transfected by hnRNP A1 or non-specific siRNA in the presence of lipofectamine 2000.

Result: OSCC tissues showed significantly higher expression of hnRNP A1 than normal tissues (p<0.01), indicating hnRNP A1 may be associated with OSCC. Knockdown of hnRNP A1 significantly retarded Cal27 cell growth, indicating hnRNP A1 is required for the proliferation of OSCC cells.

Conclusion: hnRNP A1 is overexpressed in OSCC and required for the growth of OSCC cells.
