Correlation of 3-D biofilm structure with varying resin composite topographies

Objectives: To determine correlations between the 3-dimensional structures of Streptococcus mutans biofilms with varying resin-composite topographies produced by polishing.  Methods: Specimens of Point 4 (Kerr Corp.) resin-composite were sequentially prepared (180-1200grit SiC-disks followed by polycrystalline-diamond suspensions) with final polishes equivalent to 5, 3, 1 or 0.5μm (dimensions d=6mm, h=3mm). Control group specimens were prepared against Mylarª strips and left unpolished. Surfaces were imaged using a confocal laser scanning microscope (CLSM) [Leica TCS-NT, reflection mode; three locations/specimen] before UV sterilization. Streptococcus mutans (strain UA159) biofilms were grown microaerophilically on specimens (batch culture, 0.5X Tryptone-yeast-extract medium with 10mM sucrose, 37°C, 100rpm; media replenished after 24h). After 48h bacterial growth, specimens were stained [Concanavalin A-Alexa Fluor 647 conjugate (AF; extracellular polymeric-substances), Sypro Red (SR; proteins), Syto 9 (SY; nucleic-acids); Invitrogen]. Biofilms were imaged using a Leica TCS-SP2-MP CLSM [transmission mode; three comparable locations/specimen]. Topographical parameters (Leica software;v2.61) calculated were: mean surface roughness (Ra), average maximum height (Rp) and average maximum depth (Rv). Biofilm structural parameters (ISA3D software) calculated were: porosity (PR), mean biomass thickness (BT) and bioroughness coefficient (BR). Parameters were analyzed for correlations with polishing groups using Pearson's Correlation, and by separate one-factor ANOVA tests per parameter (α=0.05).

Conclusions: Polishing of the resin-composite produced distinctly different topographies that demonstrated a positive correlation with 48h S. mutans biofilm structures (r ³ 0.45).
