Combination effect of oncolytic adenovirus with ethanol for human cancer cells

Objectives: Oncolytic adenovirus is an attractive tool for cancer therapy. E4orf6-deleted adenovirus, dl355, is an oncolytic adenovirus. This virus is able to replicate in cancer cells, in which AU-rich element (ARE) containing mRNA is stabilized. ARE is an RNA element that enhances the rapid decay of mRNA. HuR is an RNA binding protein which controls the fate of ARE-mRNA. This protein is involved in the export and stabilization of ARE-mRNA. In the vast majority of cancer cells, HuR is exported to transfer ARE-mRNA for its stabilization. In this study, we investigated the combination effect of adenovirus E4orf6-deleted mutant, dl355 and ethanol for both cancer and normal cells.
Methods: HeLa, A549 and BJ cells were used in this study. We examined HuR localization in the cells treated with ethanol by western blot analysis in order to detect the best concentration of ethanol. We examined the cell viability using XTT assay and cytopathic effect (CPE) assay. The virus replication was determined by staining hexon protein (Virus Titer assay).
Results: Cytoplasmic HuR expression was up-regulated by ethanol treatment in cancer cell. Cytolytic activities of the combination with dl355 and ethanol were significantly higher than those of virus or ethanol treatment alone. Furthermore, cytoplasmic HuR expression was enhanced by the combination treatment of cancer cells and also the additional number of viral replication was observed in combined therapy.
Conclusions: Our data indicate that the combination of oncolytic adenovirus and ethanol would be a good strategy for cancer therapy.
